Polladhavan follows Prabhu (Dhanush), a jobless yet well-meaning young man from a middle-class family in Chennai. He loves his father's old Yamaha RX 100 motorcycle more than anything else. When he falls in love with Hema (Divya Spandana), he sees the bike as his ticket to independence and romance.

The plot is set in motion when Prabhu's beloved bike is stolen. In his desperate search to recover it, he inadvertently gets entangled with a ruthless gangster named Sethu (Kishore) and a corrupt politician Daas (Murali Sharma). The stolen bike becomes a macabre carrier of hidden contraband, leading Prabhu into a world of violence, betrayal, and a high-stakes chase.
